DIAGNOSTIC DEVELOPMENT FOR WILD FIRE SURVEILLANCE Rev. Protección Veg.
Álvarez,Elba; Iglesia,Aleika.
Information about the characterization and diagnostic of wild fire was compiled. This bacterial disease, exotic for Cuba, is caused by Pseudomonas syringae pv. tabaci. The species Pseudomonas syringae is traditionally divided into pathovars depending on the host range and symptoms. For its study, different phenotypic and genotypic methods are used. Such methods allow the traditional taxonomic confirmation of this pathovar within the species. Serological and molecular techniques such as: ELISA tests, immunofluorescence, polymerase chain reaction (PCR) are also used for the characterization and detection of the bacterium. Such techniques are very useful and sensible for the diagnostic.

Tejada,TS; Silva,CSJ; Lopes,NA; Silva,DT; Agostinetto,A; Silva,EF; Menezes,DB; Timm,CD.
ABSTRACT Salmonella species have been isolated from various kinds of food and are accountable for outbreaks of foodborne diseases in humans. This study aimed at identifying the similarities between the DNA profiles of Salmonella isolated from chicken feces, chicken products, and human feces in southern Brazil. Six hundred samples were collected (200 from chicken products, 200 from broiler chicken feces, and 200 from human feces) and tested for the presence of Salmonella. Isolates proven to be Salmonella compatible by biochemical and serological tests were tested by the Polymerase Chain Reaction. Their DNA profiles were then analyzed by PFGE and rep-PCR. Salmonella was isolated from 16 out of 600 analyzed samples, with Schwarzengrund serotype presenting the...

Endophytic bacterial diversity in banana 'Prata Anã' (Musa spp.) roots Genet. Mol. Biol.
Souza,Suzane A.; Xavier,Adelica A.; Costa,Márcia R.; Cardoso,Acleide M.S.; Pereira,Marlon C.T.; Nietsche,Silvia.
The genetic diversity of endophytic bacteria in banana 'Prata Anã' roots was characterized. Two hundred and one endophytic bacteria were isolated, 151 of which were classified as Gram-positive and 50 as Gram-negative. No hypersensitivity response was observed in any of the isolates. The rep-PCR technique generated different molecular profiles for each primer set (REP, ERIC and BOX). Fifty readable loci were obtained and all of the fragments were polymorphic. Amplified ribosomal DNA restriction analysis (ARDRA) of the isolates based on cleavage with four restriction enzymes yielded 45 polymorphic bands and no monomorphic bands. PCR amplified the nifH gene in 24 isolates. 16S rDNA sequencing of the 201 bacterial isolates yielded 102 high-quality sequences....

Phenotypic and genotypic characterization of Salmonella Enteritidis isolates BJM
Oliveira,Sílvia Dias de; Bessa,Marjo Cadó; Santos,Luciana Ruschel dos; Cardoso,Marisa Ribeiro de Itapema; Brandelli,Adriano; Canal,Cláudio Wageck.
In order to study the epidemiology of Salmonella Enteritidis outbreaks and determine the source of contamination so that a recurrence can be avoided, detailed characterization is necessary. Thus, the purpose of this study was to verify whether rep-PCR was able to discriminate among Salmonella Enteritidis isolates. Phage typing, detection of virulence genes and antimicrobial resistance testing were also associated to rep-PCR results. One hundred and two S. Enteritidis isolates from broiler carcasses, food, human, pigs, poultry-related samples, and nine isolates from other countries were genotypically typed by REP-PCR, ERIC-PCR and BOX-PCR, collectively called rep-PCR. Phage typing, detection of virulence genes and antimicrobial resistance testing were also...

Repetitive element palindromic PCR (rep-PCR) as a genetic tool to study interspecific diversity in Euphorbiaceae family Electron. J. Biotechnol.
Rampadarath,Sillma; Puchooa,Daneshwar; Bal,Subhasisa.
Background The classification of diversity in germplasm collections is important for plant breeding. The repetitive element palindromic-polymerase chain reaction (rep-PCR) technique was used to investigate inter-specific diversity within 17 species from the Euphorbiaceae family using REP and BOX primers. Results The agglomerative cluster analysis was used to evaluate the scoring data. BOX and REP gave amplification with polymorphism of 98.84% and 100% respectively. REP marker demarcated between the subgenus peltatae. Both markers confirmed Jatropha tanjorensis as a natural hybrid between Jatropha gossypifolia and Jatropha curcas. Five random sequences from the rep-PCR gels were chosen, cloned and sequenced. The blast results demonstrated that the amplified...

Rhizobial characterization in revegetated areas after bauxite mining BJM
Borges,Wardsson Lustrino; Prin,Yves; Ducousso,Marc; Le Roux,Christine; Faria,Sergio Miana de.
Abstract Little is known regarding how the increased diversity of nitrogen-fixing bacteria contributes to the productivity and diversity of plants in complex communities. However, some authors have shown that the presence of a diverse group of nodulating bacteria is required for different plant species to coexist. A better understanding of the plant symbiotic organism diversity role in natural ecosystems can be extremely useful to define recovery strategies of environments that were degraded by human activities. This study used ARDRA, BOX-PCR fingerprinting and sequencing of the 16S rDNA gene to assess the diversity of root nodule nitrogen-fixing bacteria in former bauxite mining areas that were replanted in 1981, 1985, 1993, 1998, 2004 and 2006 and in a...

The genotypic diversity and lipase production of some thermophilic bacilli from different genera BJM
Koc,Melih; Cokmus,Cumhur; Cihan,Arzu Coleri.
Abstract Thermophilic 32 isolates and 20 reference bacilli were subjected to Rep-PCR and ITS-PCR fingerprinting for determination of their genotypic diversity, before screening lipase activities. By these methods, all the isolates and references could easily be differentiated up to subspecies level from each other. In screening assay, 11 isolates and 7 references were found to be lipase producing. Their extracellular lipase activities were measured quantitatively by incubating in both tributyrin and olive oil broths at 60 °C and pH 7.0. During the 24, 48 and 72-h period of incubation, the changes in the lipase activities, culture absorbance, wet weight of biomass and pH were all measured. The activity was determined by using pNPB in 50 mM phosphate buffer...
